Amino Acid Analysis

Question

What is the best sample preparation for Tryptophan analysis in protein hydrolysates?

Answer

Tryptophan is destroyed by hydrochloric acid during conventional acid hydrolysis. In order to preserve it, an alternative alkaline hydrolysis must be performed using sodium hydroxide. However this hydrolysis method is only suitable for tryptophan analysis.
